Design Notes
We digested the upstream PO Promoter (BBa_I746361) with EcoRI and SpeI and digested the downstream GFP plasmid (BBa_I13401) with XbaI and PstI. These digested fragments were transferred to a Chloramphenicol vector that was cut with EcoRI and PstI. This produced one intermediate that was selectively grown on a Chloramphenicol plate. This intermediate in combination with our other upstream intermediate, the Cadmium Promoter and Ogr Activator (BBa_K1042012) would create a final construct that is theoretically a cadmium sensor with increased sensitivity.
